I'm pretty sure that the exclamation mark in front of a message means that it is an urgent priority message. Although there is no way to change the priority of the SMS messages sent from a BlackBerry that I know of.
The priority of a message shouldn't inhibit the option to reply to it. Maybe you should try a battery pull?

Did you just upgrade to 4.5? When I upgraded, all my texts for a day or so had that mark by it. After that, it went back to normal.

FWIW, I noticed when I was running 4.5 that the "!" was on all the messages that were restored from a Back-Up.

It has to do more with the OS than with any issues. Almost every Beta OS I've run would have the "!" before sms messages and say reply requested. Some OS's make all sms that way, others don't.
